Product Description
RPMI 1640 Medium, no phenol red
Description
Roswell Park Memorial Institute (RPMI) 1640 medium was originally developed to culture human leukemic cells in suspension and as a monolayer. RPMI 1640 has since been found suitable for a variety of mammalian cells including HeLa, Jurkat, MCF-7, PC12, PBMC, astrocytes, and carcinomas. We offer a variety of Gibco RPMI 1640 modifications for a range of cell culture applications. Find the right formulation using the media selector tool.
This RPMI 1640 is modified as follows:
| With | Without |
| L-glutamine | Phenol Red |
The complete formulation is available.
Gibco RPMI 1640 is unique from other media because it contains the reducing agent glutathione and high concentrations of vitamins. RPMI 1640 contains biotin, vitamin B
12, and PABA which are not found in Eagle's Minimal Essential Medium or Dulbecco's Modified Eagle Medium. In addition, the vitamins inositol and choline are present in very high concentrations.
Product Intended UseFor
in vitro diagnostic use. CAUTION: Not for human or animal therapeutic use. Uses other than the intended use may be a violation of local law. Customers using Gibco RPMI 1640 in a manufacturing process, who have a submission with the FDA, may request a letter of authorization from us to reference our Type II Drug Master File (DMF).
cGMP Manufacturing and Quality SystemGibco RPMI 1640 is manufactured at a cGMP compliant facility, located in Paisley, Scotland, UK. The facility is registered with the FDA as a medical device manufacturer and is certified to the ISO 13485 standard. For supply chain continuity, we offer an identical Gibco RPMI 1640 product made in our Grand Island facility (11835-030). This facility is registered with the FDA as a medical device manufacturer and is certified to the ISO 13485 standards.
RPMI 1640 contains no proteins, lipids, or growth factors. Therefore, RPMI 1640 requires supplementation, commonly with 10% Fetal Bovine Serum (FBS). RPMI 1640 uses a sodium bicarbonate buffer system (2.0 g/L) and therefore requires a 5-10% CO
2 environment to maintain physiological pH.

FAQs of RPMI 1640 Medium, no phenol red:
Q: How should RPMI 1640 Medium, no phenol red be stored to maintain stability and sterility?
A: Store unopened bottles at 2C to 8C to preserve sterility, shelf life, and medium efficacy. Avoid repeated freeze-thaw cycles and always use aseptic techniques during handling.
Q: What kinds of cells and experiments is this medium most suitable for?
A: This medium is ideal for hybridoma, lymphocyte, and a variety of mammalian cell cultures, particularly in applications sensitive to indicator dyeslike fluorescence or absorbance-based assays.
Q: When do I need to supplement the medium, and with what components?
A: Supplement the medium as per your application requirements. Common additions include 10% fetal bovine serum (FBS), antibiotics to prevent contamination, and L-glutamine for enhanced cell metabolism.
Q: Where is RPMI 1640 Medium typically used in the research process?
A: It is primarily used in in vitro cell culture rooms or laboratories equipped with sterile hoods and incubators, supporting the growth and maintenance of cells for biological analysis.
Q: What is the process for using this medium in cell culture?
A: After equilibrating the medium to room temperature, aseptically supplement it if required. Add cells to an appropriate vessel, then introduce the medium, ensuring a sterile environment to avoid contamination.
Q: What are the benefits of using RPMI 1640 Medium without phenol red?
A: Excluding phenol red eliminates potential interference in assays sensitive to color changes, providing clearer results in fluorescence or colorimetric studies while maintaining optimum conditions for cell growth.
